ABSTRACT

Objective::To observe effect of addition and subtraction therapy of Huaihuasan combined with Taohuatang to ulcerative colitis with cold-heat complicated syndrome at active stage, and to study regulation effect to immune function and inflammatory response. Method::One hundred and twelve patients were randomly divided into control group and observation group by random number table. Patients with light and middle symptoms in control group got mesalazine slow release tablets, 1.0 g/time, 3 times/days, patients with severe symptoms or whose symptoms were not changed after getting for 4 weeks in control group got prednisone acetate tablets, 0.75 mg·kg-1·d-1 for 3 times. Based on the treatment in control group, patients in observation group added Huaihuasan combined with Taohuatang, 1 dose/day. The course of treatment was 4 weeks. At remission period, mesalazine slow release tablets were used for maintain long-term maintenance therapy, 0.5 g/times, 3 times/days. Scores of disease activities were graded by improvement mayo, and clinical remission and clinical efficacy were recorded, scores of cold-heat complicated syndrome, mucous membrane under enteroscopy and histology of mucosa belongs to Geboes were graded. And levels of tumor necrosis factor-α(TNF-α) in peripheral blood, interleukin-8 (IL-8), IL-10, T lymphocyte subsets (CD4+, CD8+), and adverse reactions, 6 months' follow-up and recurrence were also be recorded. Result::Therapeutic effect of traditional Chinese medicine syndromes were analyzed by rank sum test, which in observation group was better than that in control group (Z=1.915, P<0.05). Clinical effect in observation group was 98.04%(50/51) higher than 84.00%(42/50) in control group, clinical remission rate was 94.12%(48/51) higher than 78.00%(39/50) in control group, and mucosal healing rate was 96.08%(49/51) higher than 82.00%(41/50) in control group (P<0.05). Scores of mayo, cold-heat complicated syndrome, colonic mucosa and index scores of Geboes were all lower than those in control group (P<0.01). And levels of TNF-α, IL-8 and CD8+ were lower than those in control group (P<0.01), and levels of IL-10, CD4+ and CD4+ /CD8+ were higher than those in control group (P<0.01). Recurrence rate during 6 months in observation group was 10.42%(5/48) lower than 30.77%(12/39) in control group (χ2=5.669, P<0.05), as for adverse reactions, there was no significant difference between two groups. Conclusion::Huaihuasan combined with Taohuatang can induce UC to remission period, inhibit the activity of disease, alleviate clinical symptoms, regulate immune function and expression of inflammatory factors, alleviate inflammatory reaction, promote intestinal mucosal healing, and can maintain clinical remission and reduce recurrence. The clinical efficacy is superior to that of 5-ASA/glucocorticoid in Western medicine.

ABSTRACT

Objective@#Interferon-induced transmembrane protein 3 (IFITM3) is an important member of the IFITM family. However, the molecular mechanisms underlying its antiviral action have not been completely elucidated. Recent studies on IFITM3, particularly those focused on innate antiviral defense mechanisms, have shown that IFITM3 affects the body's adaptive immune response. The aim of this study was to determine the contribution of IFITM3 proteins to immune control of influenza infection .@*Methods@#We performed proteomics, flow cytometry, and immunohistochemistry analysis and used bioinformatics tools to systematically compare and analyze the differences in natural killer (NK) cell numbers, their activation, and their immune function in the lungs of -/- and wild-type mice.@*Results@#-/- mice developed more severe inflammation and apoptotic responses compared to wild-type mice. Moreover, the NK cell activation was higher in the lungs of -/- mice during acute influenza infection.@*Conclusions@#Based on our results, we speculate that the NK cells are more readily activated in the absence of IFITM3, increasing mortality in -/- mice.

ABSTRACT

Objective:To observe the effects of addition and subtraction therapy of Danggui Shaoyaosan combined with Huaihuasan on ulcerative colitis (UC) with syndrome of dampness-heat in large intestine during active stage, and the effects on brain-gut petide neurotransmitter and inflammatory cytokines. Method:A total of 130 cases were included and randomly divided into control group and observation group, 65 cases in each group. In the control group, the patients received oral administration of mesalazine enteric-coated tablets, 1.0 g/time and 3 times/days. Severe patients received prednisone acetate tablets (0.75 mg·kg-1·d-1) in several times by oral administration. Based on the treatment in control group, patients in observation group also received addition and subtraction therapy of Danggui Shaoyaosan combined with Huaihuasan, 1 dose/day. Both groups were treated for 4 weeks. Symptom scores, Mayo scores, colonic mucosa scores and Inflammatory Bowel Disease Questionnaire (IBDQ) scores were assessed before and after treatment. Patients in remission stage were followed up for 6 months to record the recurrence. Before and after treatment, vasoactive intestinal peptide (VIP), substance P (SP), somatostatin (SS), interleukin-1 (IL-1), IL-6, IL-4 and IL-10 were detected. Result:After 4 weeks of treatment, the clinical remission rate was 93.22%in the observation group, better than 80.7%in the control group (χ2=4.035,PPPPχ2=4.509,PPPPPPConclusion:On the basis of conventional western medicine treatment, addition and subtraction therapy of Danggui Shaoyaosan combined with Huaihuasan in the treatment of UC (dampness-heat in large intestine) during active stage can control the disease activity in a short term, promote restoration of the colonic mucosa. And delay the recurrence in a long term, reduce the recurrence rate, regulate ghrelin neurotransmitters and pro-and anti-inflammatory cytokines levels.

Objective@#To analyze the effects of the degree and location of intracranial vertebral artery(VA) lesions on the hemodynamic parameters of extracranial VA.@*Methods@#A total of 275 consecutive patients who were diagnosed as posterior circulation ischemic stroke or transient ischemic attack (TIA) with unilateral intracranial VA stenosis or occlusion in the Department of Neurology and Neurosurgery of Capital Medical University Xuanwu Hospital from January 2015 to December 2017 were enrolled. All patients were examined by head and neck vascular ultrasound, CT angiography (CTA) and/or digital subtraction angiography (DSA) within one week. According to the results of DSA or CTA, the patients were divided into mild stenosis group(53 patients), moderate stenosis group(62 patients), severe stenosis group(58 patients) and occlusion group(102 patients). The inner diameter (D), peak systolic velocity (PSV), end diastolic velocity (EDV), and resistance index (RI) of the extracranial segment (V2 segment) of the VA were recorded and analyzed.@*Results@#The PSV and EDV in the severe stenosis group and the occlusion group were significantly lower than those in the mild stenosis group and the moderate stenosis group (P=0.000), and the PSV and EDV in the occlusion group were significantly lower than those in the severe stenosis group[ (31±10) cm/s vs (46±12)cm/s, (5±4)cm/s vs (15±7)cm/s; all P=0.000], RI was significantly higher than the other three groups (0.85±0.12, 0.70±0.10, 0.66±0.07, 0.64±0.06, respectively; all P=0.000); RI in the severe stenosis group were not significantly different from those in the mild to moderate stenosis groups (P=0.044, 0.223). There were no significant differences in the inner diameter, PSV, EDV and RI between the subgroups in the severe stenosis group before or after the PICA (posterior inferior cerebellar artery)(P=0.130, 0.322, 0.865, 0.227). However, the EDV decreased and RI increased in the occlusive subgroup before the PICA when compared the subgroup after the PICA (all P=0.000).@*Conclusions@#The location and degree of intracranial VA lesions directly affect the changes of blood flow velocity and vascular resistance of extracranial VA, and the changes of low-speed and high-resistance hemodynamics of extracranial VA may indicate the existence of occlusive lesions in intracranial VA.

ABSTRACT

Objectives: To investigate the potential mechanism of cathepsins S inhibitor (CatS-I) affected ischemia-induced neovascularization in experimental mice. Methods: 8 week-old wild type (C57/BL6) mice were randomly divided into 2 groups: Control group, the mice received basic diet with intraperitoneal injection of 5% carboxymethylcellulose sodium and CatS-I group, the mice received basic diet with intraperitoneal injection of CatS-I (1mg/kg·d); all animals were treated for 17 days. n=20 in each group. Hind limb ischemia model was established at 3 days after injection in both groups. Blood flow was measured by laser Doppler blood flow analyzer, the ratio of ischemic area to non-ischemic area was calculated; protein expressions of peroxidase proliferation activated receptors-γ (PPAR-γ), p-Akt, p-eNOS and VEGF were examined by Western blot analysis at day 4 after the operation; frozen section of ischemic skeletal muscle was taken at 7 days after operation to measure capillary density by immunohistochemistry.Results: ① CatS-I inhibited blood flow recovery. Compared with Control group, CatS-I group had slower blood flow recovery in ischemic hind limb, P<0.05. ② CatS-I inhibited capillary formation. At 14 days after operation, capillary formation in non-ischemic skeletal muscles was similar between 2 groups, P>0.05; while in ischemic skeletal muscles, capillary density was lower in CatS-I group than Control group, P<0.01. ③ Compared with Control group, CatS-I group showed decreased protein expressions of PPAR-γ, p-Akt, p-Enos and VEGF, P<0.05. Conclusions: CatS-I regulated ischemia-induced neovascularization might be related to PPAR-γ activation and PI3K/Akt/eNOS signaling pathway in experimental mice.

ABSTRACT

AIM:To investigate the effect of microRNA(miR)-381-3p on neuronal injury in the hippocampus of depressive rats and the possible regulatory mechanism.METHODS:The rat model of depression was established by sub-cutaneous injection of corticosterone and the model rats received fluoxetine treatment.The body weight,open field test and biochemical indexes were measured for judging the therapeutic effect of fluoxetine.The expression of miR-381-3p,brain-derived neurotrophic factor(BNDF),Bcl-2 and Bax in the hippocampus was determined.The cultured neonatal rat hipp-ocampal neurons were pre-transfected with miR-381-3p inhibitor and then incubated with corticosterone.The cell viability, and the expression of miR-381-3p, BNDF, Bcl-2 and Bax were detected.The targeted regulatory role of miR-381-3p in BDNF was verified by luciferase reporter assay.RESULTS:Compared with depression group,fluoxetine increased the body weight,the number of cross-field activities and the content of norepinephrine,inhibited the expression of miR-381-3p and promoted the expression of BNDF in the hippocampus.miR-381-3p silencing reversed the effect of corticosterone,resulting in the increase in the survival rate of hippocampal neurons, upregulation of BDNF and Bcl-2, and downregulation of Bax expression.miR-381-3p targeted the regulation of BNDF expression.CONCLUSION: Silencing miR-381-3p protects rat hippocampal neurons from corticosterone injury,and its mechanism may be related to the upregulation of BNDF expression.

ABSTRACT

Objective To explore the correlation of serum HCV-RNA level with liver function indices and blood routine parameters in patients with hepatitis C,and provide the references for the early diagnosis and monitoring the prognosis of hepatitis C.Methods The study population comprised 651 patients who were diagnosed with hepatitis C in the Second Affiliated Hospital of Medical College of Xi' an Jiaotong University.Serum level of HCV RNA was determined by quantitative realtime polymerase chain reaction (real-time PCR).The data obtained were divided into six groups according to the result of HCV RNA level.Then the correlation of serum HCV-RNA level with liver function indices and blood routine parameters was analyzed.Results With the rising level of serum HCV-RNA,the levels of liver function indices including ALT,AST,AST/ALT,GGT and TBIL were all significant increased in hepatitis C patients.However,A/G presented a declining trend with the increase of serum HCV-RNA level.The lowest value and highest value of ALT,AST,AST/ALT,GGT,TBIL and A/G were 32.3±9.7 U/L and 96.2±13.6 U/L,31.1±8.38 U/L and 113.5±15.9 U/L,0.86±0.09 and 1.19±0.11,29.2± 14.5 U/L and 52.7±16.2 U/L,17.2±4.32 μmol/L and 26.0±5.58 μmol/L,0.98±0.07 and 1.35±0.14,respectively.Significant differences in ALT,AST,AST/ALT,GGT,TBIL and A/G were observed among these groups (F value was 457.1,656.4,149.1,40.18,46.56 and 146.98,respectively.all P<0.01).Moreover,the levels of blood routine parameters including WBC,NEUT%,RBC,H b and PLT were all significant decreased while that of LY% was increased with the rising level of serum HCV-RNA in hepatitis C patients.The lowest value and highest value of WBC,NEUT%,LY%,RBC,Hb and PLT were (4.30±0.22) ×109/L and (6.02±0.27) × 109/L,(48.13±3.56)% and (59.28±3.40)%,(31.05±3.41) % and (38.81 ± 4.65)%,(3.73 ± 1.70) × 1012/L and (4.65± 1.88) × 1012/L,(122.01±5.58) g/L and (135.37 ±8.50)g/L,(102.65± 16.87) × 109/L and (148.21 ± 14.40) × 109/L,respectively.There was no significant difference in RBC among groups (F=1.926,all P<0.05).However,the differences in other blood routine parameters than RBC among groups were statistically significant (F value of WBC,NEUT%,LY%,Hb and PLT was 349.0,132.145,43.65,38.91 and 52.21,respectively.all P<0.01).Conclusion Serum HCV-RNA level was correlated with routine liver function indices and blood routine parameters.It is important to perform the combined detection of serum HCV-RNA level,routine liver function indices and blood routine parameters in hepatitis C patients,which can help to guide diagnosis and to evaluate the prognosis.

ABSTRACT

Objective To observe the effect ofYinqiao Powder on the mouse models with upper respiratory trace mucosal immunity dysfunction infected with influenza virus A, and explore mechanism of action.Methods The mouse models of upper respiratory trace mucosal immunity dysfunction induced by cold stimulation with the influenza A/PR/8/34 (H1N1) virus through the nasal cavity were established. The mice were randomly divided into normal group, model group, positive medicine (Ribavirin) group, andYinqiao Powder group. All administration groups received gavage with relevant medicine, and then mortality, the life prolonging rate, average survival time and the lung index of each group were observed.Results Compared with the model group, the mortalities in positive medicine group andYinqiao Powder group decreased significantly (P<0.05,P<0.01), with longer survival time. The lung indexes in positive medicine group andYinqiao Powder group decreased significantly (P<0.05,P<0.01), and the inhibition ratios of lung index were 35.5% and 24.6%, respectively.ConclusionYinqiao Powder can realize the protective effects on upper respiratory infection through upregulating the mucosal immunity of the upper respiratory tract of mouse models.

OBJECTIVE@#To explore the effect of okra extract on gestational diabetes mellitus (GDM) rats and its probable molecular mechanism.@*METHODS@#A total of 30 female SD rats were caged with male rats for pregnancy, 27 pregnant rats were obtained and weighed. The pregnant rats were equally randomized into the control group, GDM group and intervention group. Once the pregnancy was verified, GDM group and intervention group were given 45 mg/kg streptozotocin by peritoneal injection for inducing GDM, control group was given equal volume of citrate buffer. Once the model was established successfully, intervention group was administered orally the solution containing 200 mg/kg/d okra extract, the other groups were given the diet and water only. On the 19th day of pregnancy, the blood samples and fetal rats of all groups were collected, fetal rats weight and placental weight was recorded and the serum glucose, lipids, serum insulin and C-peptide of pregnant rats before the delivery were determined.@*RESULTS@#The pregnant rats weight before the delivery, fetal rats weight and placental weight of GDM group were lower than control group and intervention group (P  0.05). Antioxidant enzymes levels of GDM group in liver and pancreas tissues were lower than the other groups, and after treatment of okra extract, antioxidant enzymes levels in liver and pancreas tissues were equivalent to control group (P > 0.05).@*CONCLUSIONS@#Okra extract, rich in antioxidant substances, could avoid the excessive consuming of antioxidant enzymes, then, suppresses the oxidative stress and insulin resistance, thereby improving blood glucose level of GDM rats.

ABSTRACT

Objective To study the clinical characteristics of neonates with Listeriosis sepsis and explore the nursing experience. Methods The clinical data of 22 cases with neonatal Listeriosis sepsis were retrospectively analyzed.Results Twenty-two neonates developed Listeria monocytogenes sepsis 0.5 h~5 d after birth with 13 cases of low birth weight. The stay was 2~77 d,10 were discharged after recovery,7 were discharged after signature of their families due to improvements and 5 died of meningitis and septicemia.Conclusion Timely and accurate collection of samples should be done for laboratory examinations.Close observation of the diseases,attaching importance to infant feeding and implementation of infant developmental nursing are all critical for the improvement of cure rate.

Objective To compare the levels of indoleamine 2,3-dioxygenase (IDO) in the peripheral blood mononuclear cells(PBMCs) from HIV-1 infected and HIV-1 negative individuals and in human tumor cells in the presence or absence of TLR ligand stimulation.Methods TaqMan probe real-time RT-PCR method for human IDO mRNA was established; IDO mRNA levels in the PBMCs from HIV-1+ and HIV-1-individuals were tested; IDO mnRNA levels in mucosal origin(T84,Caco-2,Hela) and leukocyte origin(THP-1,MT-4) tumor cells before and after exposure to agonists for TLR4,TLR7/8 and TLR9 were examined.Results It was found that a high level of IDO mRNA could be found in HIV-1+ individuals( 103.42 copy IDO mRNA/106 copy GAPDH mRNA) ; however,some high risk HIV-1-individuals may have also a high level of IDO mRNA.Some of the tumor cells could express higher level of IDO mRNA after exposure to TLR agonist.Conclusion This study indicated a role for IDO in the viral persistence and tumor formation in HIV/AIDS and further studies were warranted.

Objective To explore the compound matrine injection on human hepatoma smmc-7721 cells' proliferation.Methods Human hepatoma smmc-7721 cells were sterilely cultured.Assayed the effects of compound matrine injection on smmc-7721 cells by MTT.The concentrations of the compound matrine injection were 3.34,2.38,1.43 and 0.48 mg/ml; Used the double staining method of annexin V-fluorescein isothiocyanate(Annexin V-FITC)/propidium iodide (propidium iodide,PI) to measure the apoptosis of hepatoma cell.The concentrations of the compound matrine injection were 0.86,1.72,and 3.43 mg/ml.Results The apoptosis rates of compound matrine injection with different concentration of 3.34,2.38,1.43 and 0.48 mg/ml on liver cancer cell SMMC-7721 were51.03%、53.67%、49.83% and 0.03% respectively after 24 hours; and 67.14%、65.35%、62.25% and 0.05% respectively after 48 hours;and 74.16%、68.77%、66.04% and 26.02%respectively after 72 hours.The apoptosis rates of compound matrine injection with different concentration of 0.86,1.72 and 3.43 mg/ml on liver cancer cell SMMC-7721 were(2.86±0.35)%,(15.16± 1.15)%and(13.17±0.40)% respectively after 24 hours; and (8.57±0.44)%,(28.07±0.76)% and (29.81±8.10)%respectively after 48 hours,which were significantly higher than the control group (1.05±0.09)%(P<0.05).Conclusion The compound matrine injection has a significant inhibition on human hepatoma smmc-7721 cells' proliferation and the mechanism was its inducing cell apoptosis.

ABSTRACT

<p><b>OBJECTIVE</b>To compare the differences in the therapeutic effect on irritable bowel syndrome (IBS) between acupuncture at Tianshu (ST 25) and Dachangshu (BL 25) and western medication with Trimebutine Maleate.</p><p><b>METHODS</b>Forty cases were divided randomly into an acupuncture group and a western medication group, 20 cases in each one. In acupuncture group, acupuncture was applied to Tianshu (ST 25) and Dachangshu (BL 25). Ziwu Daojiu needling technique was adopted, once daily. In western medication group, Trimebutine Maleate capsule was administered, 2 capsules in each time, 3 times per day. The assessment on the therapeutic effect was performed in 4 weeks of treatment in two groups.</p><p><b>RESULTS</b>As compared with those before treatment, the time of abdominal pain, the frequency of abdominal pain, the morbidity of abnormal stool appearance, the morbidity of defecation abnormality, the morbidity of mucus stool and the score of bloating or abdominal pain on bowel movement were all reduced after treatment in two groups (all P < 0.01). The results in acupuncture group were much more significant than those in western medication group (the total score: 16.70 +/- 2.40 vs 15.70 +/- 3.01, P < 0.01). The total effective rate in acupuncture group was 95.0% (19/20), which was superior to that of 70.0% (14/20) in western medication group (P < 0.05).</p><p><b>CONCLUSION</b>Acupuncture at Tianshu (ST 25) and Dachangshu (BL 25) may remarkably relieve the clinical symptoms of IBS and its efficacy is superior to that of oral medication with Trimebutine Maleate.</p>

ABSTRACT

<p><b>OBJECTIVE</b>The aim of this study was to estimate and predict the AIDS epidemic situation in Jiangsu province by using a software named Estimation and Projection Package (EPP) and then to provide accurate information for making control plans and conducting interventions.</p><p><b>METHODS</b>Date were obtained from the comprehensively monitored sentinels of AIDS and venereal diseases (Data were from 28 national monitored stations and 52 provincial stations. From 2003 to 2009, a total of 10 730 000 people had been monitored) as well as project survey and laboratory tests. EPP epidemic model was employed to analyze the prevalence and to predict future epidemic situations.</p><p><b>RESULTS</b>A total of 830 000 and 1 020 000 people were monitored in 2003 and 2004 respectively, and 8 880 000 more people were monitored in the following 5 years. By the end of 2009, a total of 4103 HIV infection cases had been reported, among which 918 people were diagnosed with AIDS and 432 died. Since 2003, the cases infected through IDU had decreased from 66.84% (262/392) to 16.40% (142/868). However, the cases infected through sex contact had increased from 21.68% (85/392) to 77.40% (672/868). Among these cases, homosexual transmission and heterosexual transmission accounted for 39.10% (339/868) and 38.30% (333/868) respectively. It was estimated that there would be 14 290 HIV/AIDS cases in 2011 which is 18.10% (2191/12 099) higher than that in 2009. The estimated HIV infection rate would be 0.02%.</p><p><b>CONCLUSION</b>EPP prediction showed the AIDS epidemic situation in Jiangsu province had an ascendant trend and the AIDS epidemic situation had demonstrated rapid growth. Therefore, it is necessary to take effective preventive measures to control the spread of AIDS.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To explore the effects of survivin antisense RNA on apoptosis and chemosensitivity to docetaxel in gastric cancer line SGC7901 cells, and its relation to mdr-1.</p><p><b>METHODS</b>survivin antisense eukaryotic vector anti-pcDNA3-svv was transfected into SGC7901 cells by electorporation and positive clone was screened out. survivin protein and mdr-1 mRNA were determined by Western blot and RT-PCR. Apoptosis-inducing effect was examined by electron microscopy. Sensitivity to docetaxel was examined by MTT. Expression of mdr-1 and survivin mRNA were detected in the SGC7901 cells after drug-resisitance induction.</p><p><b>RESULTS</b>The expression of survivin protein of SGC7901 cells after transfection reduced significantly than that of non-transfected cells. MDR indexes of transfection group and non-transfection group were 0.196 +/- 0.013 and 3.126 +/- 0.019, respectively. The IC50 of transfection group to docetaxel was (16.7 +/- 1.98) microg/L and non-transfection group was (55.7 +/- 1. 89) microg/L, with a statistically significant difference. Expression of survivin mRNA in drug-resistant cells decreased along with the decreasing of mdr-1.</p><p><b>CONCLUSION</b>Antisense surivivin RNA can induce apoptosis in gastric cancer cells and increase sensitivity to docetaxel. The reversing mechanism of drug resistance is related with decreasing of mdr-1.</p>

ABSTRACT

Hepatic fibrosis is the most important pathologic course for chronic hepatic injury developing into hepatic cirrhosis. It is meaningful for treating liver disease by inhibiting the development of hepatic fibrosis. Recently, herbs of promoting blood circulation for removing blood stasis has their unique characteristics and exhibits great therapeutic effect on hepatic fibrosis. The review explains the function and mechanism of the promoting blood circulation for removing blood stasis herbs in treating hepatic fibrosis from the aspects of the monomer, effective constituent and compound recipe. It provides basis for application of promoting blood circulation for removing blood stasis herbs in study and treating hepatic fibrosis.
